1.0 PURPOSE:
1.1 To lay down the procedure for operation of polarimeter.
2.0 SCOPE:
2.1 This SOP is applicable for polarimeter in Quality control department.

5.0 PROCEDURE:
5.1 Connect the instrument to 230 volts A/C power supply.
5.2 Ensure the lid is closed.
5.3 Switch on the instrument.
5.4 Instrument initializing is displayed in the screen.
5.5 After initialization of instrument select the lamp.
5.5.1 Select and Press Na/Hal or Hg button, automatically lamp will be on. Wait for 10 minutes (Lamp warming).
5.6 Select Apeture:
5.6.1 Press the apret button and select the Normal aperture.
5.7 Select rotation mode:
5.7.1 Press the Mode button and select the SROT mode by using ?,? keys and press enter button.
5.8 Set concentration:
5.8.1 Press the Conc button and enter the concentration in g/100 mL by using numerical keys as per standard test procedure and press the Enter button.
5.9 Set the path length:
5.9.1 Press the Path button and enter the cell length by using numerical buttons and press the Enter button.
5.10 Select wavelength:
5.10.1 Press the Wavlen button and enter the wavelength by using ?,? buttons. Select the wavelength and press the Enter button.
5.11 Set the integration time:
5.11.1 Press the Integ button and enter the integration time by using numerical buttons and press the Enter button.
5.12 After setting all parameters press Set button.
5.13 Identification of the sample
5.13.1 Enter the sample particulars like sample No., sample ID comment of the sample and sample temperature by using computer keypad.
5.13.2 Enter the recorder range by using ?,?buttons.
5.14 Clean the cell with the blank solvent.
5.15 Fill the cell with the blank solvent and close with stopper.
5.16 Then clean outside of the cell with the blank solvent
5.17 Wipe the cell with tissue paper.
5.18 Place the cell in sample compartment and close the lid.
5.19 Automatically measure the optical rotation
5.20 Press the Zero button to set the optical rotation reading on to zero.
5.21 Rinse the cell thoroughly with sample solution.
5.22 Fill the cell with the sample solution and close with stopper.
5.23 Follow the points 5.16 to 5.19.
5.24 Specific optical rotation of the sample displays on the screen.
5.25 Press the Print button and collect the report and data enter in to a polarimeter logbook.
5.26 Specific optical rotation of the sample is displayed on the screen
5.27 Switch off the instrument after completion of analysis.
5.28 Clean the cell with the blank solution and finally with methanol.
5.29 Store the cell in the holder
