To lay down the procedure for operation of SHIMADZU-1601 UV-Visible spectrophotometer.
1.0 PURPOSE:
1.1 To lay down the procedure for operation of SHIMADZU-1601 UV-Visible spectrophotometer.
2.0 SCOPE:
2.1 This procedure is applicable in Quality Control Department (Block-D).

5.0 PROCEDURE:
5.1 Photometric:
5.1.1 Open the LCD unit.
5.1.2 Put on the switch at left back corner of the instrument.
5.1.3 LCD unit displays initialization and after initialization it displays mode. Select 1 for photometric.
5.1.4 In the keyboard, press go to WL to set wavelength required.
5.1.5 Rinse the cells and fill with solvent. Wipe the transparent sides of the cells with lint free filter paper.
5.1.6 Insert the cells in the cell compartments.
5.1.7 Close the shutter of the compartment and press the key AUTO ZERO on the keyboard. It will adjust to zero.
5.1.8 Remove the cell from the sample cell compartment, rinse with sample, fill with sample and place in its place.
5.1.9 The LCD display shows the absorbance at the desired wavelength.
5.2 Spectrum:
5.2.1 Open the LCD unit.
5.2.2 Put on the switch at left back corner of the instrument.
5.2.3 LCD unit displays initialization and after initialization it displays mode. Select 2 for spectrum.
5.2.4 Set the desired parameters in the spectrum window.
5.2.5 Rinse the cells and fill with solvent. Wipe the transparent sides of the cells with lint free filter paper.
5.2.6 Insert the cells in the cell compartments.
5.2.7 On keyboard press the key F1. (For base line correction).
5.2.8 Remove the cell from sample compartment, rinse with sample, fill with sample and place in its place. Then press start.
5.2.9 LCD displays spectrum.
5.2.10 After completing the spectrum, press F2. (For peak data). From this, select 3 for peak details.
