Description

All the above liquid media should be incubated for 48 hours at 32.5 ± 2.5 0C before use. Any contaminated media should be discarded

   PREPARATION PROCEDURE

  1. Weigh 58.09 g of media in a butter paper using a clean weighing balance.
  2. Transfer the weighed media into clean bottle / beaker  containing 1000 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 6.9 ± 0.2.
  4. Heat to dissolve the medium completely by keeping the beaker / bottle on the hot plate.
  5. Sterilize the media bottle in an autoclave at 121–1240C for 30 minutes.
  6. Avoid over heating of the media. 
  7. After sterilization cool the media to 40 – 500C.
  8. Dispense about 15 to 20 ml of the media into Petri plates under Laminar Airflow. Allow it to solidify.

Purpose

  1. Brilliant green agar is recommended for selective identification of Salmonella species.

 Storage condition

  1.  Hygroscopic. Store in a cool dry place below 300C away from bright light.

  2. Transfer the weighed media into clean Media bottle containing 1000 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 7.6 ± 0.2.
  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Sterilize the media bottle in an autoclave at 121–1240C for 30 minutes.
  6. Avoid over heating of the media. 
  7. After sterilization cool the media to 40 – 500C.
  8. Dispense about 15 to 20 ml of the media into Petri plates under Laminar Airflow. Allow it to solidify.

Purpose

  1. Bismuth sulphite agar is recommended for selective identification of Salmonella species.

Storage condition

  1.  Hygroscopic. Store in a cool dry place below 300C away from bright light.

  PREPARATION PROCEDURE

  1. Weigh 63 g of media in a butter paper using a clean weighing balance.
  2. Transfer the weighed media into clean Media bottle containing 950 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 7.0 ± 0.2.
  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Sterilize the media bottle in an Autoclave at 121–1240C for 30 minutes.
  6. After sterilization cool the media to 40 – 500C.
  7. Aseptically add 50 ml of Conc. Egg yolk emulsion (FD045) and 3 ml of    3.5 % Sterile potassium tellurite or 50 ml of Egg yolk tellurite Emultion.
  8. Mix well and Dispense about 15 to 20 ml of the media into Petri plates under Laminar Airflow. Allow it to solidify.

Purpose

  1. Baired parker agar is recommended for selective identification of Staphylococci.

 Storage condition

  1.  Hygroscopic. Store in a cool dry place below 300C away from bright  light.

  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Boil to dissolve the medium. Do not Autoclave and avoid over heating.
  6. After boiling cool the media to 40 – 500C.
  7. Dispense about 15 to 20 ml of the media into Petri plates under Laminar Airflow. Allow it to solidify.

Purpose

  1. Deoxycholate citrate agar is recommended for selective identification of Salmonella species.

 Storage condition

  1.  Hygroscopic. Store in a cool dry place below 250C away from bright light.

Purpose

  1. Tetrathionate broth is recommended as selective enrichment medium for Salmonella species.

        PREPARATION PROCEDURE

  1. Weigh 65 g of media in a butter paper using a clean weighing balance.
  2. Transfer the weighed media into clean Media bottle containing 1000 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 7.4 ± 0.2.
  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Dispense as required and Sterilize the media bottle in an Autoclave at  121-1240C for  30 minutes.

Purpose

  1. TSI agar is recommended as confirmatory medium.

  2. Transfer the weighed media into clean Media bottle containing 1000 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 7.2 ± 0.2.
  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Dispense as required and Sterilize the media bottle in an Autoclave at 121–1240C for 30 minutes.
  6. After sterilization cool add 20 ml of 1% sterile Potassium tellurite medium and  distribute the medium into petriplates.

Purpose

  1. V J agar is recommended as selective identification medium for Staphylococcus aureus.

  1. Weigh 56.68 g of media in a butter paper using a clean weighing balance.
  2. Transfer the weighed media into clean Media bottle containing 1000 ml of distilled water/ Purified water.
  3. Adjust the final pH (at 250C) 7.4 ± 0.2.
  4. Heat to dissolve the medium completely by keeping bottle on the hot plate.
  5. Heat to boil with frequent agitation . Dispense as required and Sterilize the media bottle in an Autoclave at 121–1240C for 30 minutes.
  6.  Do not over heat
  7. After sterilization cool the media to 40 – 500C and distribute the medium into petriplates.

Purpose

  1. XLD agar is recommended as selective identification medium for Salmonella species.
