3.0 Responsibility:
3.1 The responsibility of operating the Microscope as per procedure lies with Microbiologist.

5.0 Procedure :
5.1 Place a slide over the stage, specimen side up and center of the specimen to be examined as accurately as possible over the hole in the center of the stage with the help of slide mover.
5.2 Adjust the light source until it passes the maximum amount of light through the specimen with the lower power of objective in position, lower the body tube by means of the coarse adjustment until the objective is visible.
5.3 Look at the image through the eyepiece and focus on the specimen with the fine adjustment knob.
5.4After examining the specimen with low power objective, shift to the high power objective by rotating the nosepiece until the objective clicks into the microscopic field diameter under magnification.
5.5Look through the eyepiece and slowly raise the body with the coarse adjustment until the specimen comes into the approximate focus, then bring the image into final ocular focus by using the fine adjustment knob. Once it is set, adjust the light source and its diaphragm to give the clearest possible image.
5.6 While using oil immersion objective lens (100X), first put small drop of cedar wood oil on the specimen and touch the objective lens with oil very slowly with the help of coarse adjustment knob than focus the specimen by rotating fine adjustment knob.
5.7 Precautions:
5.7.1 Keep the microscope and lens system clean.
5.7.2 Never touch the lenses. Wipe the lens with a tissue paper gently.
5.7.3Never leave a slide on the microscope, when it is not in use.
5.7.4Always remove oil from the oil immersion lens after its use with a lens paper, which is slightly moistened with xylene.
5.7.5Keep the stage clean and dry.
5.7.6Not to tilt the microscope when working with oil immersion system.
5.7.7When the microscope is not in use, keep it in the case provided.
