Operation Of Jasco P 1030 Polarimeter

1.0    PURPOSE:

1.1    To lay down the procedure for operation of JASCO–P–1030 POLARIMETER.

2.0    SCOPE:

2.1    This procedure is applicable for Quality Control Department (Block-D).

5.0    PROCEDURE FOR MEASURING OPTICAL ROTATION AND SPECIFIC OPTICAL ROTATION:

5.1    Switch on the main instrument, which is placed left corner of the rear side. switch on the computer.
5.2    Double click on JASCO – P – 1030 Icon on the desktop.
5.3    A Spectra Manager window will appear on the screen.
5.4    Double click on standard menu.  A initialization dialogue box will appear on the screen. Initialization takes about 5 minutes.
5.5    Once the initialization is completed, the spectra manager window appears, another window also appears with the message “Optical Rotation”.

5.6    Minimize the spectra manager window.  Enter the sample identifications in optical rotation window.
5.7    Rinse the polarimeter cell with the blank solution well and fill it with the same and keep it properly inside of the cell chamber that is in front side of the Instrument.
5.8    In optical rotation window, click measure and then click Blank.
5.9    Another window appears.  Click and select enable blank.  Click measure blank.
5.10    The window is inactivated during the blank measurement.
5.11    Once the blank measurement is over, the window becomes activated again.
5.12    Click O.K. The blank value is displayed in the Optical Rotation Window.
5.13    Remove the cell.  Discard the blank solution.  Rinse well with the sample solution.  Fill the cell with same solution and keep it in the chamber.
5.14    Go to measure menu and click parameters.  The measurement parameters window appears.
5.15    Enter the parameters as per STP. In the measure mode field, select the variable to be measured e.g. Optical rotation or specific optical rotation.  Click Save.  A save as window appears.  Fill the name and click save and then click O.K.
5.16    A dialogue box appears with the message transferring measurement parameters and then this will automatically disappear.
5.17    Optical Rotation window will appears again.  Click data and click New.  Click measure and then click start.  The selected variable in the measure mode field e.g. Optical rotation or specific optical rotation is automatically measured and the results are displayed.
5.18    Take the print out of the result report.  Close the file.  Switch off the instrument and switch off the PC also.  Remove the cell from chamber and discard the sample solution.  Wash it well with Milli-Q water and replace it properly in its place.

6.0    PROCEDURE FOR CHANGING THE WAVELENGTH:

6.1    For different wavelengths, remove the magnetic lid on the top of the instrument when the instrument is in off condition.  Then remove the present filter in it and replace is with the required filter.
6.2    Now switch on the instrument and PC software.  Double click on JASCO – P – 1030 Icon on the desktop.
6.3    A spectra manager window will appear on the screen.
6.4    Click on Environment present in spectra manager box.
6.5    An initialization dialogue box will appear on the screen.  Initialization takes about 5 minutes.
6.6    An environment P- 1030 / A 010660639-dialogue box appears.  Click on 2nd source Hg.  Click O.K.
6.7    Then proceed as per steps 5.6 to 5.18 of operating procedure.
