1.0 PURPOSE:
1.1 To lay down a procedure for operation of FTIR spectrometer (Jasco).
2.0 SCOPE:
2.1 This SOP is applicable for operation of FTIR spectrometer (Jasco) in Quality control department.

5.0 PROCEDURE:
5.1 Connect the instrument to 230 Volts A/c power supply and switch on it.
5.2 Switch on the resume button located on the backside of the instrument.
5.3 Then switch on the power button, located on the top of the instrument.
5.4 Wait for 10 minutes for the initialization of lamp.
5.5 Switch on the computer.
5.6 Double click on the JASCO FT/IR – 410 icon.
5.7 Spectra manager shall be displayed.
5.8 Double click on the spectra measurement.
5.9 Spectra measurement window display on the screen.
5.10 In tool bar open measure and click background +Parameters.
5.11 Parameters/Back ground measurement screen shall be displayed.
5.12 Click standard, enter resolution as 4cm-1 and scan times.
5.13 Select Transmittance % mode and single back ground in vertical axes.
5.14 Click “Interferometer”, select gain, scan speed and filter as auto mode.
5.15 Enter the vaporization function as cosine in FFT/ times and click ok.
5.16 The background spectrum shall be scanned.
5.17 Open measure and click background.
5.18 After completion of background scanning, spectra analysis screen shall be displayed.
5.19 Prepare a pellet of the sample using KBr press.
5.20 Place the pellet in the sample holder.
5.21 Open measure menu and click sample + parameters.
5.22 Repeat 5.12 to 5.15
5.23 The scanning sample spectrum shall be displayed.
5.24 Open measure menu and click sample.
5.25 After completion of sample scanning, spectra analysis screen shall be displayed.
5.26 Click processing, select correction and adjust the baseline of spectrum.
5.27 Click other, select comment to enter the sample identification.
5.28 In peak process enter the noise level and execute.
5.29 Open view menu, select scale to adjust the scale of the spectrum.
5.30 In peak mode, select Bar X to display the wave number with peaks.
5.31 Click ”file” and select overlay to compare the sample spectrum with standard.
5.32 Save the file with a file name.
5.33 Connect a suitable printer to the system, enter print title and select “Print ” to print the Spectrum of sample and enter in to a FTIR logbook.
5.34 After completion of analysis switch off the instrument.
