Cleaning and Operation of Autoclave for Decontamination

Description

1.0    OBJECTIVE:

To lay down the procedure for cleaning and operation of Autoclave used for Decontamination.

2.0    SCOPE:

    This SOP is applicable to cleaning, and operation, of Autoclave used for Decontamination in Microbiology laboratory. 

3.0    RESPONSIBILITY: 

    Microbiologist : To perform the Operation as per SOP.

Head –Quality Control : To ensure the compliance of the SOP 

4.0    PROCEDURE: 

4.1    General cleaning:

4.1.1    Clean the Autoclave with a cloth moistened in disinfectant solution.

4.1.2    Clean the Autoclave daily.

4.1.3    Record the cleaning in “Equipment Cleaning Record” 

4.2    Precautions: 

4.2.1    Check the calibration status of the pressure gauge, Chamber Sensor, Load Sensor , digital Temperature Controller .

4.2.2    Do not open the lid of Autoclave, when cycle is running. 

4.2.3    Take care during handling of the articles, as they will be hot, after sterilization.

4.2.4    After completion of cycle, open the lid of Autoclave, when the pressure gauge shows “0” reading and temperature gauge hows below 90 OC.

4.2.5    Ensure heating coils are completely submerged in  Distilled water.

4.3    Operation: 

4.3.1    Insert plug into main electric power socket. 

4.3.2Unscrew the Locking Knobs of the Autoclave.

4.3.3Open the lid of Autoclave by turning top lifting level in Anticlockwise direction.

4.3.4    Close the Drain Valve.

4.3.5     Check the water level, water level shall be slightly above the tray.

4.3.6    Place two chemical indicators at predetermined locations.

4.3.7    Collect all contaminated plates, pipettes, used pure culcture tubes, flasks in container dedicated for destruction, close the lid of container, put the container in Autoclave, tight the Locking Knobs , select program desired programme by pressing “ UP” or “DOWN button then press “ START” button on control panel. The cycle will run for 30 minutes (As set in program No.-3).  After completion of cycle clean the Autoclave with a cloth moistened in disinfectant solution.

4.3.8            For decontamination cycle, collect all contaminated plates, pipettes, pure culcture tubes, flasks in container dedicated for destruction, close the lid of container, put the container in autoclave, tight the wing nut, select program No –3 by pressing “ UP” or “DOWN button then press “ RUN” button on control panel. The cycle will run for 30 minutes (As set in program No.-3).  After completion of cycle clean the Autoclave with a cloth moistened in disinfectant solution.

4.3.9    Record the details of the decontamination cycle.

4.4    Procedure for setting the program in program controller:

4.4.1    Switch “On” the autoclave. Set the program as given below:

4.4.2    For setting program press ENTER KEY the display will show EDIT.

4.4.3    Press again ENTER it will show SET TEMP.

4.4.4    Set the required temp (121°C) by pressing “ UP” or “DOWN” button.

4.4.5    When set temperature is displayed, press ENTER.

4.4.6    It will display  “ SETIN” Set the required time (30 minutes) by pressing “UP” or “DOWN” button .

4.4.8    Again press ENTER It will display OF SET. This value is always 0.4.

4.4.9    Press ENTER it will display INT this is always 999.9.
