1.0    PURPOSE

1.1    To lay down the procedure for Preparation of media.

2.0    SCOPE:

2.1    This SOP covers the procedure for media preparation in Microbiology Lab.

3.0    RESPONSIBILITY:

3.1     Microbiologist 

4.0    ACCOUNTABILITY:

4.1     Department Head – Quality Control

5.0    PROCEDURE:

5.1    Weigh the required quantity of dehydrated media in a clean butter paper using top pan balance as per current version of GTP210 and record the weigh details in QCxxxxx.
5.2    If the container is having new / different batch check for growth promotion test of media.
5.3    Suspend the media in a glass / Stainless Steel container containing desired volume of purified water.
5.4    Check the pH of the medium, if necessary adjust the pH by 0.1N NaOH or by 0.1N HCl.
5.5    Allot the Lot No to the each medium as ‘AA/000’.
5.5.1    Where, Alphabets AA denotes the medium code in numeric as 01,02,03 and so on and Numeric 000 denotes the Lot number (Refer appendix  –I for media codes).

5.6    For example, Nutrient agar, Lot No can be written as 15/001. 
5.7    Mix well and boil it to dissolve the medium, using a glass rod. Distribute the boiled media into glass flasks/bottles as per the requirement and record in QC0xxxxx
5.8    Keep 1 bottle with around 50ml of medium of the same Lot for pH after sterilization.
5.9    Plug each container with non-absorbent cotton and wrap each flask using butter paper with rubber band.
5.10    Use plastic cap for closing the bottles.
5.11    Load the media and operate the double autoclave to sterilize the media.
5.12    After sterilization, enter into the controlled area. 
5.13    Unload the double door autoclave.
5.14    Transfer the media bottle (which was kept for pH determination) for pH check through pass box. 
5.15    Allow to cool down the medium around 40°C for agars and room temperature for liquid medium.
5.16    Check the pH and record in QCxxxxx
5.17    If the pH is in the range as mentioned in GTP210 then, use the medium for analysis.
5.18    If the pH is not in the range discard the complete medium.
