Operation And Calibration Of Electro Lab Tapped Density Apparatus

Description

1.0    PURPOSE:

1.1    To lay down the procedure for operation and calibration of electrolab tapped density apparatus

2.0    SCOPE:

2.1    This procedure is applicable for Quality Control department.

3.0    RESPONSIBILITY:

3.2    Senior Executive – QC

5.0    OPERATING PROCEDURE:

5.1    User method:    

5.1.1    Switch on the instrument, which is at the right back side of the instrument.
5.1.2    Select user mode by pressing the left arrow key.
5.1.3    Press the set key.  Set the number of taps as per STP.  For this, select the digit pressing the left arrow key.  Then increase or decrease the digit using upward and down ward key.
5.1.4    Take known volume sample in 100 mL measuring cylinder for USP1 holder and 250 ml measuring cylinder for USP2 holder. Place measuring cylinder in the USP1 or USP2 holder according to method.
5.1.5    Press start key, tapping starts.  Once the tapping is over.  Note the volume after tapping and calculate the tapped density as per STP.

5.2    USP method:
5.2.1    Switch on the instrument, which is at the right back side of the instrument.
5.2.2    Select USP mode by pressing the left arrow key.
5.2.3    Press Method key to select USP1 or USP2 Method.  The blinking of red L.E.D. indicates selected method.
5.2.4    The number of drops is ? 300 drops/min. for USP1 and ? 250 drops/min. for USP2.
5.2.5    Use 100 mL measuring cylinder for USP1 method and 250 mL measuring cylinder for USP2 method.
5.2.6    Press the set key, display shows ‘Tap count 1’.
5.2.7    Set the number of taps as 500 by pressing the left arrow key.  Then use upward or downward arrow key respectively to increase or decrease the digit, press enter to accept.
5.2.8    Display shows ‘Tap count 2’.  Set the number of taps as 750 by pressing the left arrow key.   Then use upward or downward arrow key respectively to increase or decrease the digit, press enter to accept.
5.2.9    Display shows ‘Tap count 3’.  Set the number of taps are 1250 by pressing the left arrow key.  Then use upward or downward arrow key respectively to increase or decrease the digit, press enter to accept and press set key.
5.2.10    Press start key.  Display shows weigh sample enter weight.
5.2.11    Take known volume sample in 100 mL or 250 mL measuring cylinder as per the USP method selected and note the net weight of sample.  Place measuring cylinder in the corresponding holder as per the USP method selected.
5.2.12    Press start key.  It asks for weight of the sample.  Enter the weight by using left, up and downward arrowed keys as described in step 5.2.5, press enter key.
5.2.13    It asks for volume of sample taken.  Enter volume by using left, up and downward arrowed keys as described in step 5.2.5, press enter key.  Display shows keep the cylinder in holder and press start to run test.
5.2.14    Press start key.  Tapping starts.  Once 500 tappings are over, it asks volume.  Enter the volume after 500 taps by using left, up and downward arrowed keys as described in step 5.2.5, press enter key.  Display shows press start to continue.  Press start key.
5.2.15    Tapping starts again.  Once 750 taps are over, it asks for volume.  Enter the volume after 750 taps by using left, up and downward arrowed keys as described in step 5.2.5, press enter key. Display shows %of difference between the two volumes.
5.2.16    If the volume difference between 500 taps and 750 taps is less than 2.0%, press stop key.
5.2.17    If the volume difference between 500 taps and 750 taps is more than 2.0%, press start key.
5.2.18    Tapping starts again. Once 1250 taps are over, it asks for volume.  Enter the volume after 1250 taps by using left, up and downward arrowed keys as described in step 5.2.5, press enter key.  Display shows %of difference between the two volumes.
5.2.19    If the volume difference between 750 taps and 1250 taps is less than 2.0%, press stop key.
5.2.20    If the volume difference between 750 taps and 1250 taps is more than 2.0%, press start key.
5.2.21    In this way continue tapping until the % of difference is less             than 2.0%.
5.2.22    When the test is completed display shows start continue stop for result.  Press stop key.
5.2.23    It gives message whether to print last result.  Select yes by using the left arrow key and press enter key.  The result is printed out.
5.2.24    Press the set key to exit the operation.
5.2.25    Switch off the instrument.     
6.0    CALIBRATION PROCEDURE:

6.1    Drop height check:

6.1.1    Shall be done by service engineer.
6.1.2    Frequency once in a year.

6.2    Precision check:

6.2.1    Set 500 taps and start tapping.  Note down the time taken for 500 tapping using stopwatch.
6.2.2     Repeat step 6.2.1 for five more times and note the time for each of them.
6.2.3    Calculate the %RSD for the six readings
6.2.4    The limit is %RSD is NMT 1.0%.
6.2.5    Perform steps 6.2.1 to 6.2.3 for both USP1 and USP2 methods.
6.2.6    Record the readings in Annexure – I.

6.3    Linearity check:

6.3.1    Set 250 taps and start tapping.  Note down the time taken for 250 tapping using stopwatch.
6.3.2    Repeat step 6.3.1 by setting the tap counts 500, 750, 1000, 1250 and 1500 respectively and note the time consumed for each of them.
6.3.3    Perform steps 6.3.1 to 6.3.2 for both USP1 and USP2 methods.
6.3.4    Record the readings in Annexure – I and calculate the correlation coefficient for number of taps and time consumed.
6.3.5    The limit is NLT 0.99.

6.4    Calibration frequency:  Precision and linearity shall be done once in three months.
