1.0 PURPOSE:
1.1 To lay down a procedure for Gas chromatograph (Perkin Elmer Auto System).
2.0 SCOPE:
2.1 This SOP is applicable for Gas chromatograph (Perkin Elmer Auto System) in Quality control department.

5.0 PROCEDURE:
5.1 Connect the instrument to 230volts A/C power supply.
5.2 Switch on the instrument.
5.3 Open the door of the oven and connect the required column as per standard test procedure.
5.4 Turn the carrier gas knob
Note: Ensure that no leakage is observed near the column ends by spraying IPA solution.
5.5 Close the oven door properly.
5.6 To create a new method:
5.6.1 Press Method icon
5.6.2 Setup # Edit # Copy displays on the display screen.
5.6.3 Select Edit by using ? key.
5.6.4 Set the flow
5.6.4.1 Set the column flow
5.6.4.1.1 Press Carrier prog key
5.6.4.1.2 Enter the flow rate (in Kpa) as per standard test procedure by using numeric keys.
5.6.4.1.3 Press Enter key.
5.6.4.1.4 Set the flow rate up to required flow rate by using carrier flow knob.
5.6.4.2 Set the hydrogen, airflow at gas station.
5.6.5 Set the split ratio
5.6.5.1 Press valve key.
5.6.5.2 On the valve by using ?key.
5.6.5.3 Adjust the split by using split knob with flow meter.
5.6.6 Set the attenuation
5.6.6.1 Press the atten key
5.6.6.2 Enter the attenuation as per standard test procedure by using numeric keys.
5.6.6.3 Press Enter key
5.6.7 Enter the FID range
5.6.7.1 Press the range key.
5.6.7.2 Enter the detector range as per standard test procedure by using numeric key.
5.6.7.3 Press Enter key.
5.6.8 Set the Oven program
5.6.8.1 Press the oven prog key.
5.6.8.2 Enter the Tem 1 (Initial temperature)as per standard test procedure by using numeric keys.
5.6.8.3 Press Enter key.
5.6.8.4 Enter the Time 1 (Initial holding time) as per standard test procedure by using numeric keys.
5.6.8.5 Press Enter key.
5.6.8.6 Enter the Rate 1 (Raising temperature time per minute) as per standard test procedure by using numeric keys.
5.6.8.7 Press Enter key.
5.6.8.8 Enter the Tem 2 (Raising temperature) as per standard test procedure by using numeric keys.
5.6.8.9 Press Enter key.
5.6.8.10 Enter the Time 2 (After raising holding time) as per standard test procedure by using numeric keys.
5.6.8.11 Press Enter key.
5.6.9 Set the Injector temperature
5.6.9.1 Press the Inject temp key.
5.6.9.2 Enter the injector temperature as per standard test procedure by using numeric keys.
5.6.9.3 Press Enter key.
5.6.10 Set the Detector temperature.
5.6.10.1 Press the Inject temp key.
5.6.10.2 Enter the injector temperature as per standard test procedure by using numeric keys.
5.6.10.3 Press Enter key.
5.6.11 Press Set (?) key.
5.7 Turn the Hydrogen gas knob on, push on ignition.
5.8 Press the Detect control button, press the push set flowed by tuning the air knob slowly.
5.9 Switch on the computer
5.10 To create a new project:
5.10.1 In Millennium window select and double click the configure system.
5.10.2 Configure system window displays on the screen.
5.10.3 Select projects and click ok.
5.10.4 System/administrator – configuration manager window displays on the screen.
5.10.5 In tool bar open file, click new and select the new project
5.10.6 New project wizard window displays on the screen.
5.10.7 Enter the table space.
5.10.8 Activate the audit trail support.
5.10.9 Click next, select owner group and world, administrator.
5.10.10 Click next; select the view filters, custom fields and methods.
5.10.11 Click next, enter the project name and comments, click finish.
5.11 To load the existing project:
5.11.1 In Millennium screen double click the browse project.
5.11.2 Browse project window displays on the screen.
5.11.3 Select required project and click ok.
5.11.4 Required project window displays on the screen.
5.12 To create a new instrument method:
4.24.1 Open the required project window.
4.24.2 In project window open file, click new and select instrument method.
4.24.3 Select General.
4.24.3.1 Description – Enter the required description.
4.24.3.2 Sampling rate – 1
4.24.3.3 Units - mµ
4.24.3.4 Sampling factor-1000
4.24.3.5 Bcd polarity – (-).
4.24.4 In project window main tool bar open file, click save as
4.24.5 Save as window displays on the screen.
4.24.6 Enter the required instrument method name.
5.13 Create new processing method:
5.13.1 Open the required project window.
5.13.2 In project window open file, click new and select processing method.
5.13.3 New processing window displays on the screen..
5.13.4 Enter the processing type – LC and click ok.
5.13.5 In integration enter the integration parameters like width, threshold, minimum area and height.
5.13.6 In suitability active the calculate suitability results and enter void volume, select the proper pharmacopoeia status.
5.13.7 Select file, click save as and enter the processing method.
5.14 To create a new report method:
5.14.1 Open the required project.
5.14.2 In project window open file, click new and select report method.
5.14.3 New method/group window displays on the screen.
5.14.4 Select create new report method, click ok
5.14.5 Select individual report click next.
5.14.6 Report type- select individual and click finish.
5.14.7 Default report open, select file, click save as and enter the report method name as required.
5.15 To create a new method set:
5.15.1 Open the required project window.
5.15.2 In project window open file, click new and select method set.
5.15.3 New method set editor window displays on the screen.
5.15.4 Click yes.
5.15.5 Select required instrument method and click next.
5.15.6 Select required process method and required report method, click next.
5.15.7 Enter the method set name, comments and click save.
5.16 To acquire single run:
5.16.1 In project, select tools and click run samples.
5.16.2 Select proper method set and instrument method.
5.16.3 At chromatogram monitor screen click the right side mouse button and select the customize channels.
5.16.4 Customize channels window displays on the screen.
5.16.5 Enter the required wavelength and press apply and ok buttons.
5.16.6 Click setup, after click the setup screen shows idle.
5.16.7 Press monitor, display the base line screen.
5.16.8 After stabilization of base line, stop the run (select the inject and click the stop run).
5.16.9 Enter the method set, injection volume and run time.
5.16.10 In tool bar open inject, click inject system (inject icon) for injection.
5.16.11 After click the Inject system icon screen shows weighting for injection. Then inject the required quantity of the sample and press RUN key on the instrument
5.17 To reprocess the chromatogram:
5.17.1 Open the project window and select the channels, click update.
5.17.2 Select and double click the required file name.
5.17.3 Review window displays on the screen.
5.17.4 In review window open file menu, click open and select method set.
5.17.5 Select the appropriate method set and click “ok”.
5.17.6 Select the process and click integrate.
5.17.7 After integration completed select the file, choose save and click result.
5.17.8 Close the review window.
5.17.9 Press update icon at particular project window.
5.17.10 Select the required file name.
5.17.11 In main tool bar open tools and click print.
5.17.12 Print window displays on the screen.
5.17.13 Select appropriate report method and click ok icon.
5.17.14 After collect the report enter the GC logbook.
5.18 After analysis completed close the project window and log off the millennium and switch of the detector and computer
