1.0 PURPOSE:
1.1 To lay down a procedure for operation of Polarimeter (Jasco).
2.0 SCOPE:
2.1 This SOP is applicable for operation of polarimeter (Jasco) in Quality control department.
3.0 RESPONSIBILITY:

5.0 PROCEDURE:
5.1 Connect the instrument to 230 volts A/C power supply.
5.2 Ensure the lid is closed.
5.3 Switch on the instrument.
5.4 Instrument initializing menu is displayed.
5.5 After initialization of instrument the main menu is displayed.
5.6 Clean the cell with the blank solvent.
5.7 Fill the cell with the blank solvent and close with stopper.
5.8 Then clean outside of the cell with the blank solvent.
5.9 Wipe the cell with tissue paper.
5.10 Place the cell in cell holder and close the lid.
5.11 Press the EDIT button.
5.12 In EDIT menu; enter the cycle number, cycle time and integration time.
5.13 Press Next: enter temperature correction, auto print mode and auto message.
5.14 Press Next; enter the cell length and comment like sample particulars and press Esc.
5.15 Open and close the Lid of the instrument.
5.16 The instrument scans the blank.
5.17 Press “ZERO” button to nullify the blank solvent reading.
5.18 “ CLEARING ZERO IS PROGRESSING “ shall be displayed
5.19 Press Edit and select next; enter the sample ID, sample concentration and press Esc.
5.20 Open the lid of the instrument.
5.21 Remove the stoppers of the cell.
5.22 Rinse the cell thoroughly with sample solution.
5.23 Fill the cell with sample solution and close with stopper.
5.24 Wipe the out side the cell with the blank solvent.
5.25 Clean the cell with tissue paper.
5.26 Place the cell in the holder and close the lid.
5.27 The instrument scans the sample.
5.28 The specific optical rotation of the sample is displayed
5.29 Press “PRINT” button.
5.30 Print window displays on the screen.
5.31 Press one by one param print, title print, data print to get the print out of the result.
5.32 Switch off the instrument after taking reading enter in to a polarimeter logbook.
5.33 Clean the cell with the blank solution and finally with methanol.
5.34 Store the cell in the holder.
