1.0 PURPOSE
1.1 To lay down the procedure for calibration of micropipette by using water 22°C
2.0 SCOPE
2.1 This SOP covers the Calibration of Micropipette and is applicable in Microbiology
3.0 RESPONSIBILITY
3.1 Microbiologist
4.0 ACCOUNTABILITY
4.1 Head - Quality Control
5.0 PROCEDURE
5.1 Operation
5.1.1. Set the required volume by red colour knob rotating clockwise and anti clockwise.
5.1.2 Fix the micro tip by applying light pressure.
5.1.3 Press the Plunger and insert the micro tip in the solution to be taken.
5.1.4 Release the plunger to get the required solution.
5.1.5 Transfer the solution by pressing the plunger little more than the normal.
5.1.6 Press the dispenser knob to remove the used micro tip.
Note: Use this pipette (20 -200µl) for volume not less than 20 µl and not more than 200 µl
Note: Use this pipette (200 -1000µl) for volume not less than 200 µl and not more than 1000 µl
5.2 Calibration
5.2.1 Set the volume at 20 µl and 50 µl for micropipette (T –200).
5.2.2 Follow the operation procedure from point no 5.1.2 to 5.1.5.
5.2.3 Take the purified water at room temperature as per set volume and weigh it in a weighing bottle, which is previously dried and weighed.
5.2.4 Record the reading.
5.2.5 Follow same procedure for other set volumes as given below.
5.2.5.1 For micropipette T –200
20-µl
50-µl
5.2.5.2 For micropipette T –1000
200-µl
500-µl
5.2.6 Repeat the analysis for 5 times and take the average.
5.2.7 Calculate the volume by dividing the average weight of water with 0.9960 to get the actual volume and record the values in annexure –I
5.2.8 Acceptance criteria:
For micropipette T –200
Set volume Permitted Volume
20- 19.8-20.2 µl
50-µl 49.6-50.4
For micropipette T –1000
Set volume in Permitted Volume
200-µl 196-204 µl
500-µl 495-505 µl
Frequency - Quarterly
