Operation of Perkin Elmer UV VIS Spectrometer

Description

1.0    PURPOSE:

1.1    To lay down the procedure for operation of perkin elmer UV/VIS spectrometer.

2.0    SCOPE:

2.1    This SOP is applicable for quality control department.

3.0    RESPONSIBILITY:

3.1    Assistant  – QC 

4.0    ACCOUNTABILITY:

4.1    Executive – QC 

5.0    PROCEDURE:

5.1    Measurement of absorbance at particular wave length:

5.1.1    Switch on the instrument, which is on the top right side of the instrument.
5.1.2    Switch on the computer.
5.1.3    Double click on UV-win lab icon.
5.1.4    UV win lab explorer window appears.
5.1.5    Click on methods in folder list.
5.1.6    Select the appropriate wavelength method from the methods list and double click on it.
5.1.7    UV-win lab window appears with method name.
5.1.8    Click on data collection in the folder list.
5.1.9    Set the wavelength by entering the wavelength in left side box and clicking add>icon.  The wavelength appears in the center box.  To remove the unwanted wavelengths select the wavelength in the middle box and click on < remove icon.

5.1.10    Select the ordinate mode A and slit width 1 nm.
5.1.11    Click on sample in folder list.
5.1.12    Enter number of samples, sample ID and description of sample.
5.1.13    Click on actions in the menu bar and click start.
5.1.14    A UV win lab window appears with the message remove sample(s) and press ok to perform and auto zero.  
5.1.15    Open the sample chamber, which is at right front of the instrument, and keep the cells containing blank solution in both front and back cell holders. Close the sample chamber and click OK.
5.1.16    After some time UV win lab window appears with the message place sample in the instrument and press OK.  To begin data collection.
5.1.17    Replace blank solution in the front cell with the sample solution.  Keep it in chamber.  Close the chamber and click OK.
5.1.18    After some time, UV win lab window appears with the message all the sample sin the table have been run click OK.
5.1.19    Click on reporting in the folder list.
5.1.20    Report appears on the screen.  Select appropriate template from the list and click on it.
5.1.21    Report appears in the selected format.  Click on preview.  Report preview appears.
5.1.22    Click print icon.  Print window appears.
5.1.23    Select print hard copy and click OK.
5.1.24    Print/reprint report window appears which the message confirm printing of this report.  Click OK.  Report is printed out.
5.1.25    After completing the measurements exit the file.
5.1.26    A window appears with message modifications have been made the task must be saved before the program can exit. click OK.
5.1.27    Save task, as window appears enter the file name and description and click save.
5.1.28    Save results window appears with the message save results.  Click OK.  
5.1.29    UV win lab explorer window appears.  Exit the file.

5.2    Scanning at a particular wavelength range:

5.2.1    Switch on the instrument, which is on the top right side of the instrument.
5.2.2    Switch on the computer.
5.2.3    Double click on UV-win lab icon.
5.2.4    UV win lab explorer window appears.
5.2.5    Click on methods in folder list.
5.2.6    Select the appropriate scanning method from the methods list and double click on it.
5.2.7    UV-win lab window appears with method name.
5.2.8    Click on data collection in the folder list.
5.2.9    Enter the start and end wavelength select ordinate mode A and slit width 1 nm.  
5.2.10    Double click on data collection in folder list. Double click on instrument.  Select a scan speed appropriate to the experiment (15, 30 etc.)  And enter data interval and number of cycles as per requirement.
5.2.11    Click on sample in folder list.
5.2.12    Enter number of samples, sample ID and description of sample.
5.2.13    Click on processing in folder list.   Click on add icon and click on select peak table by clicking it.
5.2.14    Click on actions in the menu bar and click start.
5.2.15    A UV win lab window appears with the message remove sample(s) and press ok to perform and auto zero.  
5.2.16    Open the sample chamber, which is at right front of the instrument, and keep the cells containing blank solution in both front and back cell holders. Close the sample chamber and click OK.
5.2.17    After some time UV win lab window appears with the message place sample in the instrument and press OK.  To begin data collection.

5.2.19    After some time, UV win lab window appears with the message all the sample sin the table have been run click OK.

5.2.22    Report appears in the selected format.  Click on preview.  Report preview appears.
5.2.23    Click print icon.  Print window appears.
5.2.24    Select print hard copy and click OK.
5.2.25    Print/reprint report window appears which the message confirm printing of this report.  Click OK.  Report is printed out.
5.2.26    After completing the measurements exit the file.
