To lay down the procedure for operation of LC-10ATVP-SHIMADZU LIQUID CHROMATOGRAPH – Used for washing the HPLC-Columns.
1.0 PURPOSE:
1.1 To lay down the procedure for operation of LC-10ATVP-SHIMADZU LIQUID CHROMATOGRAPH – Used for washing the HPLC-Columns.
2.0 SCOPE:
2.1 This procedure is applicable for Quality Control department.
3.0 RESPONSIBILITY:

4.2 Asst. Manager – QC
5.0 PROCEDURE:
5.1 Keep the section inlet tube of the instrument in desired solvent bottle.
5.2 Connect the pump outlet to the column inlet using S.S. tubings in proper direction mentioned on the column. Keep the outlet of the column in waste beaker.
5.3 Switch on the instrument by pressing the power switch provided in front bottom –left corner of the instrument.
5.4 Press the purge key. A message PURGING LINE IS DISPLAYED. After completion of purging, again press the purge key.
5.5 Press the pump key to switch on the pump.
5.6 Press the function key. Enter a flow rate of 0.2 mL/min. using the numerical keys. Press enter key. Gradually increase the flow rate to 1.0 mL/min. in the same way as mentioned above.
5.7 Wash the column for required period. Reduce the flow rate gradually to 0.00 mL/min. as explained in step-5.
5.8 Press the pump key to switch off the pump. Switch off the instrument by pressing the power switch.
