Amlodipine is a long-acting CCB that may be used to treat chronic stable angina (exertion-related angina) and mild to moderate essential hypertension. It also inhibits vascular smooth muscle carbonic anhydrase I activity causing cellular pH increases which may be involved in regulating intracellular calcium influx through calcium channels.
PO 5 to 10 mg every day.
In Prinzmetal's angina, hypertension and in Stable angina.
Headache, peripheral oedema, fatigue, somnolence, nausea, abdominal pain, flushing, dyspepsia, palpitations, and dizziness. Rarely pruritus, rash, dyspnoea, asthenia, muscle cramps, hypotension, bradycardia
Grape fruit can significantly increase serum levels of Amlodipine and may cause toxicity.
Hypersensitivity to dihydropyridnes with Amlodipine.
Store in tightly closed container in cool location at 59° to 86°F.
