Actions

It is a moderate-spectrum antibiotic effective against a wide range of Gram+ve, and a limited range of Gram-negative organisms. Amoxicillin binds to PBP-1A (penicillin-binding protein 1A) located inside the bacterial cell well. Penicillin-sensitive transpeptidase the penicillin’s acylate C-terminal domain by opening the lactam ring. This inactivation of the enzyme prevents the formation of a cross-link of two linear peptidoglycan strands, inhibiting the third and last stage of bacterial cell wall synthesis. Cell lysis is then mediated by bacterial cell wall autolytic enzymes such as autolysins; it is possible that amoxicllin interferes with an autolysin inhibitor.

Dosage/Dosage form

  • Adult: The recommended dose range is 0.25-3g
  • Child: 25 mg/kg/day  in divided doses for every 12 hours

Used to treat bacterial infections caused by microorganisms.

Adverse effects/Side effects

Hyperactivity, agitation, dizziness, insomnia, maculopapular rash, urticaria, exfoliative dermatitis, hypersensitivity vasculitis; diarrhoea, nausea, vomiting; anaemia, thrombocytopenia, leucopenia, and agranulocytosis.

Interaction

  • Increased levels with disulfiram and probenecid.
  • Increase effects of oral anticoagulants.
  • Decreased effects with tetracyclines and chloramphenicol.

Contraindications

Caution when used during pregnancy, Hypersensitivity and lactation.
