Actions

Inhibiting the bacterial cell wall synthesis

Dosage/Dosage form

  • 900 mg as a single dose for Treponemal infections and for Streptococcal pharyngitis as IM.
  •  For the treatment of early syphilis:  1.8 g of single dose, usually as 2 injections in separate sites while as late syphilis: 1.8 g at wkly intervals for 3 consecutive weekk for Syphilis as IM.

Therapeutic uses

Used to treat acute glomerulonephritis, respiratory tract infections, rheumatic fever and chorea, rheumatic heart disease, rheumatic heart disease, syphilis and other venereal diseases.

Adverse effects/Side effects

Hypersensitivity reactions including uticaria; fever; joint pains; rashes; angioedema; haemolytic anaemia; serum sickness-like reactions; interstitial nephritis; thrombocytopenia; neutropenia; convulsions; diarrhoea; antibiotic-associated colitis.

Interaction

  • Bacteriostatic drugs e.g. tetracyclines; chloramphenicol, other antibacterials;
  • Anticoagulants.
  • Probenecid prolongs T1/2 of benzathine benzylpenicillin.

  • In very high doses had a risk of heart failure or neurotoxicity.
  • Avoid contact, skin sensitization may occur.
