Dosage/Dosage form

IV- The recommended dose is 125mg/ m2 over 90 minutes infusion, weekly for 4 weeks followed by a gap of 2 weeks.

Therapeutic uses

Used for the treatment of cancer.

Adverse effects/Side effects

Lachrymation, anaemia, neutropenia, acute diarrhoea, thrombocytopenia; sweating, abdominal cramps, hypersalivation, miosis, weakness; vomiting, nausea, alopecia and skin reactions; cardiovascular toxicity.

Interaction

  • St John's wort
  • Diuretics may increase the risks of vomiting/diarrhoea to secondary dehydration;
  • Prophylactic dexamethasone as an antiemetic may enhance lymphocytopenia;
  • Prochlorperazine may increase incidence of akathisia; antineoplastic agents (myelosuppression and diarrhoea).
  • Ketoconazole may reduce irinotecan exposure.

Contraindications

  • Pregnancy and lactation.
  • Inflammatory bowel disease, severe hepatic impairment, bowel obstruction.
