Actions

It inhibits the cGMP specific PDE5 (phosphodiesterase type 5) which is responsible for degradation of cGMP in the corpus cavernosum located around the penis. The inhibitions of (PDE5phosphodiesterase type 5) by vardenafil enhance the erectile function by increasing the amount of cGMP.

Dosage/Dosage form

The recommended dose is 10 mg; it should be taken approximately 60 min before sexual activity as PO. The maximum dose is 20 mg.

Adverse effects/Side effects

Headache, loss of hearing, heart attack, palpitation, blurred vision.

Interaction

  • Avoid consumption of alcohol while taking Vardenafil. Do not consume grapefruit, wine or grapefruit juice while taking Vardenafil. It may cause serious drug reactions.
  • Vardenafil may interact with several drugs including conivaptan, INH, imatinib, clarithromycin, anti-depressants, erythromycin, anti-fungal drugs, tamsulosin, calcium-channel blockers, nitrates, anti-arrhythmic drugs and anti-retroviral drugs.

Contraindications

Contraindicated in patients who are taking nitrate, hypersensitivity and heart disease.

Precautions

  • Co-administration of Vardenafil with nitrate (anti-anginal drugs) may result in serious hypotension.
  • Vardenafil is contraindicated in patients with history of CHF, stroke, MI, prolonged QT syndrome, seizures, hyperlipidemia, hypotension or hypertension, leukemia, hemophilia, GI bleeding, vision changes, retinitis pigmentosa and Peyronie’s disease.
  • Do not use other drugs such as alprostadil or yohimbine while taking Vardenafil.
